Ab 1555 Mitglied in der St.-Lukas Gilde, Antwerpen und möglicherweise Maler in der Werkstatt Frans Floris''. Seine eigene Werkstatt hatte einen großen Kundenstamm, darunter den Erzherzog Ernest von Österreich. Bekannt war Mostaert für seine Winterlandschaften, doch schuf er auch Genre- und Historienbilder. Radierungen nach Werken Mostaert u.a. durch Jan Sadeler, Hendrik Hondius und Hieronymus Wierix.
